چکیده

Objective: Disruption of the intestinal epithelial barrier has been shown to occur following burn injury. This process can lead translocation pathogens from gut lumen systemic circulation and distant organs thereby increasing risk for sepsis. The aim this study was examine effect amifostine (WR-2721) on bacterial in a rat injury model. Material Method: A total 27 male Wistar albino rats were divided into three groups nine. Group I control group. II III subjected third-degree burns over 30% body surface area, group administered 200 ml/kg intraperitoneally, followed by 10 ml/kg/day maintenance dose after undergoing burns. After 48 hours, tissue blood samples obtained cultured liver, spleen, mesenteric lymph nodes, cecum. Results: Blood cultures negative all groups. In group, colonization appeared only cecum, but III, found While most frequently cecum counts did not significantly differ (P = 0.298) nodes 0.418) between III. Conclusion: Amifostine alone is effective controlling associated with injuries. These results should be interpreted caution as there are number factors that affect translocation.

Protection and potentiation of nitrogen mustard cytotoxicity by WR-2721.

The radioprotective agent WR-2721 was examined for its effects on modifying the cytotoxicity of HN2 against normal and tumor cells in the AKR mouse. Quantitation was carried out by the spleen colony assay for both normal hematopoietic stem cells and AKR leukemia cells. Protection from drug toxicity and normal cell cytotoxicity was noted. Potentiation of cytotoxicity to AKR leukemia was found.

Effects of ethiofos (WR-2721) and radiation on monkey visual discrimination performance.

WR-2721 (ethiofos) is a promising protector against radiation-induced lethality and may be useful in cancer radiotherapy (Davidson et al . , 1980). However, ethiofos also produces nausea, vomiting, diarrhea, and hypotension, which implies severe behavioral consequences (Bogo et al . , 1985). We studied the effects of ethiofos on behavior in monkeys and its ability to mitigate early transient in...

A phase I trial of amifostine (WR-2721) and melphalan in children with refractory cancer.

Melphalan has a steep dose-response curve, but the use of high doses results in unacceptable myelosuppression. Strategies to circumvent this dose-limiting myelosuppression would allow for the administration of higher, more effective doses of melphalan. Amifostine (WR-2721) has been shown in preclinical studies to protect the bone marrow from the myelotoxicity of melphalan, and in clinical trial...

Adjunctive use of ethiofos (WR-2721) with free radical-generating chemotherapeutic agents in mice: new caveats for therapy.

The neurotransmitter analogue 6-hydroxydopamine has been proposed as a selective chemotherapeutic agent for peripheral neural crest tumors. It exerts its action through the generation of cytotoxic oxygen free radicals. Unfortunately, it is toxic to normal peripheral neurons as well. Ethiofos (WR-2721) is a free radical scavenger which appears to be preferentially taken up by normal cells relati...
